To better understand the mechanisms of resistance in the biotype of Plutella xylostella resistant to bendiocarb, the P450 monooxygenase-encoding genes (CYP6B5, CYP4F8 and CYP6B4) were expressed in Sf9 insect cells. Differential inducibility of these genes and their dependence on the over-expression of P450 reductase or cytochrome P450 reductase were also investigated. The highly efficient over-expression of the P450 reductase genes was achieved by the insertion of the dsRed2 red fluorescent protein gene under control of the actin 5C promoter. RT-PCR and qRT-PCR were performed using primers designed from the nucleotide sequences of the cDNA clones for CYP6B5 and CYP6B4. Cytochrome P450 CYP6B5 was expressed constitutively. Cytochrome P450 CYP6B4 was induced by the addition of the inducer, 1-hydroxybenzotriazole, and induced at a lower level than CYP6B5. CYP6B4 was also induced by piperonyl butoxide (PBO) at a low level, but the induction was not completely blocked by PBO. The highly efficient over-expression of the reductase genes was achieved by the insertion of the dsRed2 red fluorescent protein gene under control of the actin 5C promoter. The rate of increase in the total level of CYP6B5 transcripts increased over 3.5 fold after a 6-h incubation with 1 mM of 1-hydroxybenzotriazole (1-HBT). Transcripts of CYP6B4 were increased by 2.1 fold after 6 h of 1-HBT treatment. Markup Import and Markup Assist
Markup Import and Markup Assist let you import CAD data into your drawings to quickly update designs with annotations and comments. The new Markup Import feature allows you to annotate drawings directly on PDFs.
Markup Import creates the comments and annotations in the PDF as you work. You can export the PDF and easily incorporate those changes back into your CAD files.
By opening a PDF in your preferred PDF viewer, you can review and make edits that you can quickly incorporate back into your design. If you need to export the annotations back to your design, you can export a flattened version of the PDF that you can edit and re-import back into your CAD files.
